Only if you delete the game. As long as it is installed and you have inserted the original disc it should continue to work afaik. I use this out of convenience and you can store Xbox/360/One games on a cheap external drive instead of the pricey Series SX drives.
Get a couple of tb mechanical drive and that should hold just about all the older games.
